Pixels are the size in inches times the dots per inch. 300 dpi is right for anything seen at arm's length; a poster seen from across a room can be 150. 72 dpi is only for showing on a screen.

With bleed
If the art runs to the edge, the printer wants it to run past the edge too, so a sliver of white never shows where the paper's cut. That's bleed: 0.125 in on each side is usual in the US. US Letter with bleed is 8.75 × 11.25 in, or 2625 × 3375 pixels at 300 dpi. Keep words and faces at least as far inside the edge again. Your printer's spec sheet has the last word.
